The present study, undertaken to determine the potential for IFNAR2 expression during brain development, revealed that this receptor, in its mature form, is expressed in the brain of the adult rat, but is undetectable in the developing brain. However, IFNAR2 immunoreactivity became apparent in the brain as early as embryonic day 14 following cytokine-induced maturation. Immunofluorescence studies, using nuclear counterstain, revealed that the IFNAR2 expression pattern was restricted to neuronal cells. To determine the cell types in which IFNAR2 is expressed, we performed double-labeling studies to analyze for colocalization with the neuronal marker, microtubule-associated protein 2. The results indicate that IFNAR2-immunoreactive cells co-localize with markers of neuronal cells in the developing and adult rat brain. Further studies using 7-day-old rat brain and organotypic cultures further demonstrated that IFNAR2 is expressed in neuronal cells within the developing brain. Taken together, these results suggest that IFNAR2 expression is associated with neuronal cells in the brain.
